Voltage-gated sodium channel (VGSC) \u03b21 subunits are multifunctional proteins that modulate the biophysical properties and cell-surface localization of VGSC \u03b1 subunits and participate in cell\u2013cell and cell\u2013matrix adhesion, all with important implications for intracellular signal transduction, cell migration, and differentiation. Here, we report that\u00a0 \u03b21 subunits are\u00a0<em>S-<\/em>palmitoylated. Substitution of a single residue in \u03b21, Cys-162, to alanine prevented palmitoylation, reduced the level of \u03b21 polypeptides at the plasma membrane, and reduced the extent of \u03b21-regulated intramembrane proteolysis, suggesting that the plasma membrane is the site of \u03b21 proteolytic processing.\u00a0This is the first demonstration of\u00a0<em>S<\/em>-palmitoylation of a VGSC \u03b2 subunit, establishing precedence for this post-translational modification as a regulatory mechanism in this protein family.
